Reading manga and studying the style first is really important. Having an idol is also very helpful because it will help inspire your style of drawing. In manga, the anatomy is somewhat realistic but still has a sense of perfectness to it. In very popular mangas (such as OHSHC and Fruits Basket) the characters are very skinny, pale, perfect and tend to have larger eyes to help portray the innocence, youth or overall beauty of the character. Mangas also differentiate in style, so checking out a lot before setting your goal on a certain style is important. Trying out different styles when starting out is also important, but I highly recommend choosing one and sticking with it once you find your favorite. I prefer art more along the lines of realistic manga where the characters don't look perfect and differ in body shapes and faces. It's really up to you though, what ever route you choose to take with your art is obviously going to be the right choice for you. Studying regular human anatomy and the way humans move is also important to take note on. Once you make an OC or draw a character from your favorite anime/manga, try drawing them with different facial expressions and in different poses wearing and doing different things. Ditto to what Rust said. It usually depends on your style and how realistic you want it to be. Ditto to the poses part as well. To make sure the pose works, try to do it yourself, and observe how other people move so that when you draw, your characters look alive. From personal experience, to improve, is to draw all the time. If there's something not quite right, fix it. Also, I don't recommend using a cheap pen to draw with AT ALL. Darn thing spurts out a random blob of ink when I'm doing delicate lines.... Actually, beware of trying to ink in your rough sketches. Find a good reliable pen or one of those fancy professional kinds, the kind you dip in an ink bottle. Don't be afraid to show off your art either. Don't try and cram it down people's throats though (little bit guilty on that) and don't ignore any compliments. Be proud of your work, and accept criticism, not insults. Yes, there is a difference. Also a good thing to do is to check out past artworks youve drawn. It always makes me cringe when I stumble onto a sketch I drew years ago... My friend once told me to try and redraw an old drawing, and compare what youve fixed since then and what hasnt. Shes a great artist, so yeah :D I seriously could go on and on, so I'll stop rambling now. ^^"
